In the last few years many novel and alternative protein purification protocols have been designed using immobilized short-peptides or peptidomimetics as ligands for affinity chromatography. These ligands are easy to synthesize and less expensive than protein based ligands, such as monoclonal antibodies or protein A, thus enabling low-cost industrial purification processes. Many of these peptide ligands have been selected from the screening of combinatorial peptide libraries. Those peptides with affinity for the target proteins are immobilized on chromatographic supports and their performance for selective target protein purification is assessed.This chapter is structured around three parts: a) strategies used to design short-peptide and peptidomimetic affinity ligands; b) methods to immobilize those ligands on chromatographic supports and c) applications of the affinity matrices developed.
