SummaryWnt/β-catenin signaling plays a pivotal role in hair follicle morphogenesis and in regeneration during hair cycle. We showed that DHT inhibits Wnt signal-mediated transcription in dermal papilla cells (DPC) from Androgentic alopecia (AGA) patients. We studied the molecular cross-talk between the androgen and Wnt signaling pathway in dermal papilla cells and the effect on hair follicle stem cells differentiation induced by DPC. In co-culture assays we have revealed that DHT and androgen receptor (AR) signaling activate glycogen synthase kinase-3β in DPC, by down-regulating its phosphorylation in Ser-9 and up-regulating its expression. This, inhibits the canonical Wnt pathway and consecuently diminishes hair follicle stem cells (HFSC) differentiation into follicular lineage. In assays with DPC microspheres, DHT down-regulates Wnt agonist Wnt10b while up-regulates Wnt antagonist Dkk-1. We showed that impaired HFSC differentiation in AGA is mediated by androgen modulation of Wnt agonists and antagonists expression balance in DPC. In summary , Androgen inhibition of Wnt signaling in DPC would decrease DPC inductivity, and thus, impair HFSC differentiation, contributing to hair follicle miniaturization during AGA
