TRIOLEIN REDUCES MMP-1 UPREGULATION BY DERMAL FIBROBLASTS GENERATED BY ROS PRODUCTION BY UVB-IRRADIATED KERATINOCYTESCyto􀁍ine production and oxidative stress generated by ultravioletradiation 􀀤 􀀊􀀷􀀸􀀤􀀋 s􀁍in exposure are main factors of s􀁍inphotoaging. 􀀫nterleu􀁍in􀀏􀀘 􀀊􀀫L􀀏􀀘􀀋 produced by irradiated 􀁍eratinocytesis proposed to have a role in metalloproteinases 􀀊􀀯􀀯􀀲s􀀋expression activation in dermal fibroblasts. 􀀹e examined theeffect of triolein treatment of 􀀷􀀸􀀤􀀏irradiated 􀁍eratinocytes on􀀯􀀯􀀲􀀓 􀀊interstitial collagenase􀀋 expression response of dermalfibroblasts. 􀀹e assayed 􀀷􀀸􀀤􀀏irradiated 􀁍eratinocytes solublesignals, mainly 􀀫L􀀏􀀘 and reactive oxygen species 􀀊􀀴􀀱S􀀋. 􀀫L􀀏􀀘expression and 􀀴􀀱S generation 􀁙ere analyzed in 􀀷􀀸􀀤􀀏irradiated􀁍eratinocytes. 􀀯􀀯􀀲􀀓 m􀀴􀀰A expression response 􀁙as examinedin fibroblasts gro􀁙n in 􀁍eratinocytes conditioned medium. 􀀹eevaluated the effect of treating 􀁍eratinocytes 􀁙ith triolein on 􀀫L􀀏􀀘expression and 􀀴􀀱S generation in 􀁍eratinocytes, and 􀀯􀀯􀀲􀀓expression in fibroblasts. The irradiation of epidermal cells 􀁙ithsublethal 􀀷􀀸􀀤 doses increased 􀀫L􀀏􀀘 expression and 􀀴􀀱S generation.Conditioned culture medium collected from 􀁍eratinocyte s􀁙as used to culture dermal fibroblasts. 􀀯􀀯􀀲􀀓 m􀀴􀀰A upregulation􀁙as observed in fibroblasts cultured in conditioned mediumby 􀀷􀀸􀀤􀀏irradiated 􀁍eratinocytes. Triolein treatment reduced the􀀫L􀀏􀀘 expression and 􀀴􀀱S generation in 􀁍eratinocytes and thiseffect 􀁙as reflected in do􀁙nregulation of 􀀯􀀯􀀲􀀓 expression infibroblasts. Triolein reduces both the expression of 􀀫L􀀏􀀘 and􀀴􀀱S generation in irradiated 􀁍eratinocytes. 􀀫t seems to exert ananti􀀏inflammatory and anti􀀏oxidative stress effect on irradiated􀁍eratinocytes that in turn reduces 􀀯􀀯􀀲􀀓 expression in dermalfibroblasts. Collectively, these results indicate that triolein actsas a photoprotective agent.
