Resumen:
Prolonged ingestion of water contaminated with arsenic may result in the manifestations of toxicity in practically all systems of the body. Previous studies carried out by X-ray fluorescence spectrometry using synchrotron radiation (SR-μXRF, 300x300 μm) showed a high concentration of As in the renal cortex from chronically exposed rats (Pérez et al., 2006). An additional accumulation of Cu and Zn was observed. Since As accumulation, as well as of any other trace metals, even essential ones, may therefore be critical for that organ, the present study was designed to analyze the glomerular and non-glomerular distributions of As, Fe, Cu and Zn in renal cortex from male Wistar rat exposed to As. Furthermore, time-dependent alterations in Creatinine and BUN plasmatic values and histopathology were used to the renal injure assessment. The animals received drinking water containing 50 ppm As ad libitum for 60 days (control, without arsenic). Then, 2x2 mm sections in renal cortical slices (100 μm thick) was scanned with a micro-collimated white synchrotron spectrum (30x30 mm) and the elemental distribution was determined by SR-µXRF. The results indicated a peri-glomerular accumulation of As and Cu. The deleterious effects of arsenic exposure may also be seen by decreased plasmatic creatinine and BUN values and Bowman´s space. Overall, a clear time–response relationship between arsenic exposure, As-Cu peri-glomerular accumulation, and renal injury has been founded.
