DC-10 is a novel and distinctive subset of tolerogenic dendritic cell (DC) characterized tobe a potent IL-10 producer and type 1 Tregs (Tr1)-inducer. Although DC-10 and otherssubsets of Myeloid regulatory cells (Mregs) were recently found increased at the humandecidua, their role in materno-fetal tolerance maintenance are still unknown. Due todecidualization process involves immunoregulatory factors secretion by endometrialstromal cells, we focus on the impact of this process in conditioning monocytes to Mregssubsets. Thus, conditioned media (CM) of Human endometrial stromal cell line (HESC)decidualized with MPA+dbcAMP for 8 days (Dec-CM) or not (Non-dec CM) were collected.Then, isolated monocytes from peripheral blood mononuclear cells of healthy donorswere cultured with rhGM-CSF+rhIL-4 for 5 days in the absence/presence of CM.
