Liver cold static preservation in a non-heart beating (NHB) donor model in rat. The perfomance of BGP-HMP solution versus HTK solution.

Resumen:
The aim of this work was to compare two solutions for cold preservation of marginal livers for transplant. One was the BES-Gluconate-Polyethylene Glycol solution (BGP-HMP) -originally developed for hypothermic machine perfusion- against a commercially available Bretschneider´s solution (HTK). Freshly harvested livers from Male adult Sprague Dawley rats, and livers preserved (24 h at 5°C) in BGP-HMP or HTK (Custodiol®) were immediately reperfused in the ex-vivo system for functional evaluation. Non-heart beating liver (NHB) donation was performed on sedated animals by an i.v. injection (150 μL of 2 M KCl containing 150 UI of Heparin), resulting in an immediate cardiac arrest. After 45 minutes of warm ischemia, the livers were washed, removed and stored (24 h at 5°C) in preservation solution, and then, they were reperfused in ex vivo system. Six groups were designed: I- Freshly harvested livers (Control); II- Freshly harvested livers from NHB (ischemic Control), III- Preserved livers in BGP-HMP, IV- Preserved livers in HTK, V- Livers from NHB preserved in BGP-HMP and VI-Livers from NHB preserved in HTK. The results showed that portal flow rate, intrahepatic resistance and O2 consumption from group V showed best performance than VI group. Also, NHB treatment plus HTK preservation had a diminished bile flow respect to group I. However, there were no differences between controls and NHB + BGP-HMP storage. LDH and ALT released after 90 min of reperfusion showed statistical differences (p
