Sub-Lethal Dose of Shiga toxin 2 from Enterohemorrhagic Escherichia coli Affects Balance and Cerebellar Cythoarquitecture

ShigatoxinproducingEscherichiacolimaydamagethecentralnervoussystembeforeorconcomitantlytomanifestedhemolytic?uremicsyndromesymptoms.Thecerebellumisfrequentlydamagedduringthissyndrome,however,thedeleteriouseffectsofShigatoxin2hasneverbeenintegrallyreportedbyultrastructural,physiologicalandbehavioralmeans.Theaimofthisstudywastodeterminethecerebellarcompromiseafterintravenousadministrationofasub-lethaldoseofShigatoxin2bymeasuringthecerebellarblood?brainbarrierpermeability,behavioraltaskofcerebellarfunctionality(inclinedplanetest),andultrastructuralanalysis(transmissionelectronmicroscope).Intravenousadministrationofvehicle(controlgroup),sub-lethaldoseof0.5and1ηgofStx2permouseweretestedforbehavioralandultrastructuralstudies.Asetofthreeindependentexperimentswereperformedforeachstudy(n=6).Blood?brainbarrierresulteddamagedandconsequentlyitspermeabilitywassignificantlyincreased.Lowerscoresobtainedintheinclinedplanetaskdenotedpoorcerebellarfunctionalityincomparisontotheircontrols.Themostsignificantlowerscorewasobtainedafter5daysof1ηgoftoxinadministration.TransmissionelectronmicroscopemicrographsfromtheStx2-treatedgroupsshowedneuronswithaprogressiveneurodegenerativeconditioninadosedependentmanner.Assub-lethalintravenousShigatoxin2alteredthebloodbrainbarrierpermeabilityinthecerebellumthetoxinpenetratedthecerebellarparenchymaandproducedcelldamagedwithsignificantfunctionalimplicationsinthetestbalance.
