Amniotic fluid (AF) is essential for normal fetal growth and developmentand aquaporins (AQPs) may be crucial in its regulation.To date, four AQPs (AQP1, AQP3, AQP8, AQP9) were describedin human amnion and emerging evidence showed that an alteredexpression of these proteins might be associated with abnormal AFvolume such as oligohydramnios or polyhydramnios. However, theetiologies of these syndromes are unknown.Recently, we demonstrated that AQPs facilitate water transportacross the human amnion, being the AQP1 the most important contributor.However, the effect of osmotic stress on the regulation ofthese proteins was not studied yet.Our aim was to study the effect of hypo and hyperosmolar stress onthe expression and function of the human amnion AQPs.This study was approved by the ethics committee of the Hospital NacionalDr. Prof. A. Posadas. Human amnion explants were culturedin hypo (150 mOsm) and hyperosmolar (400 mOsm) conditions generatedby dilution of the culture medium or addition of a sucrosesolution, respectively. Net transepithelial water movements acrossthe human amnion were measured using a modified Ussing chamber.AQP1, AQP3, AQP8 and AQP9 expressions were assessed byWestern Blot and semiquantitative RT-PCR.Hyperosmolar condition compared to isoosmolar condition, showeda significantly decreased in AQP1 and AQP9 expressions (n=7;p
