Diglycosidases are quite rare enzymes and occur in some bacteria,[1] fungi[2,3] and plants. They hydrolyze the heterosidic linkage of diglycoconjugates such as rutin or hesperidin, generating a disaccharide and the corresponding aglycon as products. Some diglycosidases exhibit transglycosylation activities, which provide access to various novel diglycoconjugates. We report on the cloning of several diglycosidase-encoding genes from three fungal strains: Mucor circinelloides, Penicillium chrysogenum and Acremonium sp. We succeeded in heterologously expressing some of these diglycosidases in active form using Escherichia coli or Pichia pastoris as a host. This enabled us to characterize these enzymes in terms of their substrate specificities and transglycosylation activities.
