IN SILICO STRUCTURAL CHARACTERIZATION OF TRICHODERMA KONINGIOPSIS ECH42 GENE INVOLVED IN BIOCONTROL OF PHYTOPATHOGENIC FUNGI.

BACKGROUND:Governments of different countries, as well as farmers and consumers of agricultural products, are becoming increasingly aware of the intensive application of chemicals for diseases control on crops. In this scenario, they are trying to find alternative, effective, eco-friendly and harmless to human methods. The use of microorganisms or their products (such as enzymes) as biocontrol agents is a sustainable alternative for this problem. Trichoderma koningiopsis strain POS7 is an isolate native from Misiones province, which showed a high in vitro ability to control against several phytopathogens. The characterization of genes involved in biocontrol and the identification of their response elements or putative inducers may be the key to ensure the effectiveness in the sustainable management of plant pathogens. The main objective of the present work was to structurally characterize the ech42 gene, which codes for an endochitinase responsible in the biocontrol of phytopathogenic fungi.RESULTSA consensus sequence of the ech42 gene was generated from sequences of Trichoderma species and related genera available in NCBI system. The consensus sequence was used to map the 7,773,936 reads obtained from the sequencing of the T. koningiopsis POS7 genome using the Geneious 9.1.5 software. A structural region of 1468 bp was obtained, being identified 4 exons, 3 introns, and a regulatory region of 709 bp. This last region has 2 TATA boxes, 3 CAAT boxes and 3 inverted CAAT boxes. In addition, several response elements were identified: 2 ones corresponding to physiological stress (CCCCT), 3 ones related to carbon catabolic repression (Cre1 and CreA), 3 ones to nitrogen repression (GATA), and 4 putative binding sites for proteins with regulatory functions during parasitism (MYC1, MYC2, MYC3 and MYC4).CONLUSIONSIn silico characterization of the ech42 gene of the strain T. koningiopsis POS7 that codes for one endochitinase is reported, being identified several structural features compatible with other enzymes with a putative role against phytopathogenic fungi.
