Plants are defending themselves continuously from the attack of pathogens; the production of reactive oxygen species (ROS) is one of themain defense mechanisms triggered in response to pathogens. Polyamines are aliphatic amines of low molecular weight that show animportant regulatory activity in plant cells. The higher polyamines spermidine and spermine are catabolized by polyamine oxidases (PAO) withthe concomitant production of hydrogen hydroxide (H2O2). Herein we present the functional characterization of the AtPAO1 and AtPAO2genes, which encode cytoplasmic and peroximosal enzymes, during the interaction with Pseudomonas syringae pv. tomato DC3000. Thesepolyamine oxidases mutant lines showed disturbed contents of ROS (H2O2 and superoxide anion radical). Accumulation of superoxide anionradical in Atpao mutants can be explained in part by changes in AtRBOHD and AtRBOHF transcript levels and AtRBOH activity. In addition,transcriptional regulation of AtPAO2 gene by H2O2 and Pseudomonas is presented. Our data highlight the importance of PAOs in plantdefense and their interrelation with other enzymes involved in ROS production and detoxification.
