Resumen:
Central nervous system invasion by bacteria of the genus Brucella results in an inflammatory disorder called neurobrucellosis. Blood-brain barrier activation is a common feature of human neurobrucellosis, but the underlying mechanisms are largely unknown. The aim of this work was investigated the influence of B. abortus activated-platelets (PTL) on the Human Brain Microvasculature Endothelial Cells (HBMEC). For this, we stimulated human PTL with B. abortus. Supernatants (SN) were used to stimulate HBMEC. Expression of ICAM-1 and secretion of IL-6, IL-8 and MCP-1 were evaluated on HBMEC. Sn of B.a. stimulated-platelets activated HBMEC as we observed a higher ICAM-1 expression and a higher secretion of IL-6, IL-8 and MCP-1 (p0.05), demonstrating that a soluble factor was implicated on the HBMEC activation. The viability of B.a. was not necessary, as SN of Heat kill B.a. stimulated PTL activated HBMEC. Soluble CD40L was not implicated on the mechanism, as an anti-CD40L antibody could not blocked the activation of HBMEC. Treatment of SN with trypsin avoid the effect (p
