Resumen:
Fermentation patterns of Escherichia coli HBlOl carryingplasmids expressing cloned genes of Zymomonas mobilispyruvate decarboxylase (PDC) and alcoholdehydrogenase II(ADH) were determined in glucose-limitedcomplex mediumin pH-controlled anaerobic batch cultivations. Timeprofilesof glucose, dry cellweight, succinate, formate, acetate, andethanol were determined, as were the activities of ADH andPDC. Fluxes through the central carbon pathways were calculatedfor each construct utilizing exponential phase dataon extracellular components and assumingquasi-steadystate for intermediate metabolites. Overallbiomass yieldswere greatest for cells expressing both PDCand ADH activities.Yields of carbon catabolite end products were similarfor all PDC-expressing strains anddifferent from thosefor other strains. Relative to its glucoseuptake rate, thestrain with greatest PDC and ADHactivities produces formateand acetate more slowly and ethanol morerapidlythan other strains. Strong influences of plasmidpresenceand metabolic coupling complicate detailedinterpretationsof thedata.
