Escherichia coli HBlOl was grown in complex medium under anaerobic andaerobicconditions. Cells prepared under these twodifferent conditions were characterized bytwo-dimensional protein gelelectrophoresis, by NMR measurements under identical(anaerobic) conditions, and by measuringthe kinetics of glucose uptake and cataboliteend-product appearance in the medium underidentical anaerobic conditions. Specificrates of glucose uptake and end-productformation were significantly greater for theanaerobically grown cells, which alsoexhibited lower intracellular concentrations of sugarphosphates, nucleoside di- andtriphosphates, UDPG, and NAD(H). Two-dimensionalgel electrophoretic analyses reveal changesin the intracellular levels of proteins involvedin pyruvate catabolism that have beenobserved previously for E.coli grown in minimalmedium under aerobic and anaerobicconditions. Enzymes involved in the TCA cyclewere not detected in cells grown aerobically or anaerobicallyin complex medium.
