Resumen:
IGF2BP1 is an RNA-binding protein highly expressed in various types of solid tumors. Its exp ression was found to promote HCC cell proliferation, migration, invasion and correlate with poor survival and prognosis. Sorafenib (Sfb) is the standard systemic therapy used for advanced HCC. Resistance to Sfb still remains one of the major causes of treatment failure and often involvesthe upregulation of ABC transporters, like ABCB1 and ABCC3. We aim to evaluate whether IGF2BP1 is involved in ABCB1 and ABCC3 modulation by Sfb in HCC cell lines. HepG2 and HuH7 cells were incubated with Sfb 2μM for 48h and 72h; DMSO was added to control cells (C). IGF2BP1 was transiently knocked down with shRNA targeting human IGF2BP1 mRNA (sh-IGF2BP1) orscrambled (sh-SCR) as control in HepG2 cells.
