Decidualization process involves phenotype and functional changes on endometrial cells and the modulation of different mediators such as cytokines, chemokines and the selective recruitment of immune cells. The vasoactive intestinal peptide (VIP) is an immunoregulatory neuropeptide produced by endometrial stromal cells. Here we explored the possible immunomodulatory role of VIP during decidualization.Human endometrial stromal cell line (HESC) was treated or not with VIP or with medroxiprogesterone (MPA) and dbcAMP. VIP/VPAC system and decidualization markers were evaluated by RTPCR and ELISA. Invasion assay was performed using blastocyst-like spheroids (BLS) from first trimester trophoblast cells (Swan71) cultured on HESC monolayer. The activation profile of dendritic cells (DC) and monocytes, and the migration of monocytes and Treg cells were evaluated by flow cytometry. Foxp3 expression was evaluated on C57BL/6 wildtype or VIP-deficient mice uterus.Decidualization increased VIP secretion in HESC cells (p
