Resumen:
Dendritic cells are capable to present antigens from the extracellular microenvironment associated to molecules of MHC class I, triggering the activation of CD8+ lymphocytes. Previously, we demonstrated that histamine (HIS) enhances the cross-presentation of soluble ovalbumin (OVA) from murine dendritic cells (DCs). Taking to account that cross presentation involved 3 pathways (vacuolar, retrograde and phagocytic), the aim of this study was to analyze the possible mechanisms responsible for this increase. For test this point, we assayed the expression of MHC class I-OVA after the treatment of DCs obtained from bone marrow precursors with Epoximicin (Epo;5µM) or Bafilomycin A1 (Baf;5µM); proteosome and ATPase inhibitors, respectively. Interestingly, OVA presentation through MHC class I was only affected by HIS (1µM) in DCs pre-treated with BAF (Ct: 2.2 ± 00.7; HIS: 3.3 ± 0.9; CtBaf: 1.7±0.5; HISBaf: 1.1±0.4, % positive DCs ± SE; *p
