Resumen:
Objectives: During the decidualization process, a subset of endometrial stromal cells (EnSCs) undergoes cellular senescence. Wehypothesize that recurrent implantation failure (RIF) patients displayan imbalance between both subpopulations. Since these patientshave been reported to show alterations in endoplasmic reticulumstress (ERS), we studied the link between both senescence andERS using an in vitro model. Materials and methods: Deiodinase2 (DIO2), Lumican (LUM) and Clusterin (CLU) were evaluated asmarkers for the senescent subset, while Ferritin (FTL) was usedto evaluate the decidual one. Forkhead box protein O1 (FOXO1)was tested as a key transcription factor involved in the divergenceof the subpopulations. Gene expression was tested in endometrialbiopsies from fertile women, RIF patients and Human EndometrialStromal Cell line (HESC) by RT-qPCR. To evaluate how ERS affectssenescence in EnSCs, HESC cells were ERS induced using 1μg/mlof thapsigargin (Tg) for 4h. Then, stimulus was washed, and supernatants and RNA were recovered after 48 h *p
