Resumen:
Adverse pregnancy outcome and deep placentation disorders areassociated with periodontal disease. Porphyromonas gingivalis(Pg), one of the main pathogens causing periodontitis, releasesextracellular vesicles known as outer membrane vesicles (PgOMV)that play an important role in the host-pathogen interaction. During placentation, trophoblast cells (Tb) secrete soluble factors in a closeinteraction with maternal leukocytes to maintain immune homeostasis. An impaired trophoblast-immune interaction is associated withpregnancy complications. Previously we reported that PgOMV diminished the migration and invasion capacity of Tb. Our aim wasto study the effect of PgOMV on Tb cell metabolism and trophoblast-immune interaction. Human first trimester trophoblast cell lineHTR-8 was stimulated with 1 µg/ml PgOMV for 4-24h. Lipid droplets, glucose and fatty acid uptake were analyzed by flow cytometryusing Bodipy 493/503, 2-NBDG and FLC12 probes, respectively.Lactate production was measured by Accutrend Plus system andmRNA expression of inflammatory mediators by qPCR. Peripheralblood monocytes and neutrophils were purified from healthy donorsand cultured with conditioned media from Tb (CM) pre-treated ornot with Pg-OMV (PgOMV-CM). Reactive oxygen species (ROS)production was evaluated by flow cytometry.
