Specific and differential immune responses elicited by DNA vaccines against Bovine Herpes Virus 1 (BoHV-1) vehicularized in rationally designed liposomes

Genetic vaccination is a promising alternative method to use against BoHV-1. In addition to their safety, the de novo synthesis of the proteins of immunological interest allows their presentation in the context of MHC class I and II molecules. As a result, a CTL response important in achieving good levels of protection is induced against viruses like BoHV-1.However, the low transfection efficiency of DNA vaccines is a serious concern. In order to address this issue, we used rationally designed liposomes combined with different plasmid constructions. In some formulations cell penetrating peptide (TATp) was added.Various liposomes were formulated (PLAIN-Ls and TATp-Ls) as vehicles for two vectors encoding for secreted (gDS) and the anchored forms of glycoprotein D (gDA). Vaccines were made with gDS or a mix of gDS plus gDA (gDS+A) and were intradermally inoculated in BALB/c mice.Here, we show that only gDS+A plasmids vehicularized with liposomes stimulated IFN-ã secretion, indicating a good Th1 polarization. Whereas naked (NKD) gDS was the best at eliciting humoral responses, TATp-Ls did not elicit a humoral response. In addition, only PLAIN-Ls induced lymphoproliferation when stimulated with recombinant gD (gDr). Finally, only TATp-Ls:gDS+A elicited strong cytotoxicity, followed by TATp-Ls:gDS with weakly induced cytotoxicity, using BoHV-1 infected target cells. In conclusion, these results show that the induced immune response depends both on the plasmid constructions and on the vehicle used, and that TATp-Ls are an effective mean of enhancing the induction of CTLs in vivo.
