Resumen:
Peroxiredoxins (PRDXs) are the main antioxidant defenses against oxidative stress in human spermatozoa. Prdx6-/- mouse spermatozoa have poor reproductive outcomes. Our aim was to elucidate the impact of the inhibition of 2-Cys PRDXs or the lack of PRDX6 on fertilizing competence of mouse spermatozoa.C57Bl/6J wild type (WT) mice spermatozoa were incubated in BWW medium with MJ33 (PRDX6 PLA2 activity inhibitor), or thiostrepton (TSP; 2-Cys PRDX peroxidase activity inhibitor) for 2h at 37°C. Sperm motility was measured by CASA. Embryo development, zona binding, sperm fusion were evaluated after IVF with mature oocytes. Sperm viability, lipid peroxidation, and mitochondrial superoxide (O2─) levels were determined by flow cytometry using Sytox Green, BODIPY C11 and MitoSOX Red, respectively.
