ABSTRACTThe aim of this work was to evaluate
the ability of human polymorphonuclear
leukocytes (PMNL)
to modulate the allostimulatory ability of dendritic cells (DCs). Previous report showed that PMNL increase DCs-mediated
allogenic T cell proliferation, perhaps through DC-SIGN molecule on DCs. In
this work we demonstrate that T cells stimulatory capacity of DCs incubated
with PMNL, varied in accordance to the amount of PMNL used. Proliferation of
allogeneic T cells was evaluated by coincubating DCs with syngeneic PMNLs in a
[H3] thymidine incorporation assay. DCs-induced T cell proliferation
decreased in the presence of 50.103 or 100.103 PMNL.
However, 5.103 PMNL per well increased the DCs-induced T cell
proliferation (Table). Furthermore, the expression of DC-SIGN on the surface of
DCs was evaluated in an immunofluorescent assay. A 48% decrease in MFI was seen
on DCs incubated with culture supernatant derived from 1.106 PMNLs ,
but not lower. These results suggest that PMNL are able to up or downmodulate
the allostimulatory ability of DCs, probably by modifying the expression of
DC-SIGN.
