This chapter will emphasise on the currently available methodologies that whereas allowing a proper characterisation of the photosynthetic response of leaves to water stress are non-destructive and fast to implement. These indirect methods deal basically with the spectral signature of transmitted or reflected light and the chlorophyll fluorescence emitted by leaves. Whenever possible examples will be provided for durum wheat. Other methodological approaches, mostly concerning the canopy or the whole plant levels are detailed elsewhere.
