Elicitation of Rubia tinctorium cultures with methyljasmonate to increase anthraquinone production

Plants produce a defense response against elicitation that involves the production of secondary metabolites, such as anthraquinones (AQs), and the generation of reactive oxygen species (ROS). One of the agents widely used is methyl jasmonate (MeJ). AQs biosynthesis involves different metabolic routes: áketoglutarate and isochorismic acid (IC) are precursors of A and B rings. IC is produced from chorismic acid, the end-product of shikimate pathway (SP). The terpenoid pathway originates the C ring. The pentose phosphate pathway (PPP), which generates erithrose-4-phosphate (substrate of the SP), has been propose to be coupled to the proline cycle: the NADP+ generated by this cycle could act as cofactor of the first enzymes of the PPP. The aim of this work was to study the relationship between MeJ elicitation, ROS, proline cycle and AQs production in R. tinctorum. Cell cultures were treated with MeJ, diphenyl iodonium (DPI, an inhibitor of H2O2 generation) and with glucose/glucose oxidase (Glu/GOD, a H2O2 generating-system). MeJ increased AQs content (53%) after 48 hs. DPI addition reduced AQs accumulation in MeJ treated cells at 24 (67%) and 48 hs of culture (49%). Glu/GOD induced AQs production (14 and 48%) after 24 and 48 hs. MeJ treatment increased proline content at 48 hs of culture (122%), while it was decreased in DPI/MeJ treatment (72 %). Involvement of ROS is under study.
