Here we report a study of nitrocytochrome c that combines two colors resonant raman and other biophysical techniques showing that nitration of Tyr74 induces Tyr deprotonation, which is coupled to Met/Lys axial ligand exchange. The structural change results in altered electron shuttling capability, augmented peroxidatic activity, electric field sensitivity and lipid peroxidation efficiency of nitrocytochrome c. Based on these results we analyzed the mechanism in which nitration of Tyr74 may be implicated in early events of apoptosis inducing cardiolipin lipoperoxidation.
