Glyceraldehide-3-phosphate dehydrogenase (GAPDH), one of the most abundant proteins in the cytoplasm, is considered a moonlighting protein due to its diverse functions in the cell in addition to its enzymatic activity in the glycolytic pathway. This functional diversity exhibited by GAPDH arises from its ability to bind to multiple partners. Among these partners, there are several proteins that are related to neurodegenerative diseases, such as amyloid-β peptide, Tau, α-synuclein, hungtintin. The role for these interactions is still unclear since there are reports showing that GAPDH might play a deleterious or protective effect in neurodegenerative diseases. Our results show that these seemingly contradictory roles could be understood in terms of the conformational plasticity of GAPDH. We demonstrate that under diverse conditions GAPDH forms a variety of high molecular weight aggregates that expose unique surface properties able to interact with specific target proteins in different ways. These results might help understand the role played by GAPDH in neurodegenerative diseases.
