Abstract Proteolytic enzymes of plasminogen activator system participate in several physiologic process that require a focalized and controlled effect. It was demonstrated that porcine oviductal fluid shows urokinase  plasminogen activator (u-PA) activity and the u-PA gene expression in oviductal cells. In order to know if u-PA is present in the oviduct of other not-related species we analyzed the expression of u-PA gene in vicugna oviduct. A primary cell culture of oviductal epithelial cells was used to obtain total RNA by the SV RNA Isolation System kit. Specific primers designed from the porcine u-PA cDNA sequence were used in the RT-PCR reactions. The amplified fragment of about 400 bp had a similar size to the obtained from porcine cDNA (434 bp). The nucleotide sequence analysis revealed that it presents a 87% of identity with porcine u-PA and 84% with bovine u-PA. These findings confirm that this fragment corresponds to vicugna u-PA. The nucleotide sequence codes to the carboxy terminal region of the protein and a part of the un-translated 3´ region. The presence of the u-PA mRNA in the vicugna epithelial cells indicates that u-PA is synthesized and probably secreted to the oviductal lumen. u-PA would act during the gamete transport, fertilization and/or early embryo development.
