Resumen:
Sphingolipids
are structural components of biomembranes involved in cell signaling, owing to
the interaction with specific lipids, the ability of modify the bilayer order
degree and induce segregation of or even change of phase state upon enzymatic modification.
However, a detailed characterization of the behavior of pure molecular species
and of their mixtures has not yet been done. In this work, sphingomyelins and
ceramides with different lengths and unsaturation degree of the N-acyl chain
were studied by means of Langmuir monolayers in order to assess the mean
molecular area, the resultant perpendicular dipole moment and the lateral
compressibility of the pure species and their binary mixtures as a function of
the lateral pressure and composition. The results indicate that in mixtures of
sphingomylelins and ceramides, favorable molecular interactions exist leading
to the formation of monolayers with decreased phase-transition pressures and
lateral compressibility. The study of the lateral topography of the films by
Brewster angle microscopy confirmed that the sphyngomyelin-ceramide mixtures
showed more condensed states than the pure components when compared at the same
surface pressures
