Differentially methylated DNA sequences from tomato reciprocal hybrids Gimenez, Magalí1; Cambiaso, Vladimir1,2, Rodríguez, Gustavo R.1,21Instituto de Investigaciones en Ciencias Agrarias de Rosario (IICAR-CONICET-UNR). 2Cátedra de Genética, Facultad de Ciencias Agrarias, Universidad Nacional de Rosario. The parental genotypes of those hybrids were Gema FCA and Querubín FCA, Caimanta and LA722 of Solanum pimpinellifolium and Red Purple and Zebra Green respectively. Additionally, it was determined the occurrence of genomic imprinting between the reciprocal hybrids throughout Methylation Sensitive Amplified Polymorphism (MSAP) analysis at mature green stage fruits. This analysis revealed 10 epiloci differentially methylated among reciprocal hybrids. In order to know the DNA sequence in which these epiloci, the bands were extracted from the acrylamide gels, amplified by PCR and sequenced. The obtained sequences were analyzed using BLAST at the Sol Genomics Network (https://www.solgenomics.net/tools/blast/). The 10 epiloci belong to sequences present in chromosomes 3, 4, 6, 8, 10 and 12 of the tomato genome. Six of them presented homology with coding regions of the genes IPT9 - IPT2, At1g72480, ETO1, GMI1, DEGP2 and TRZ1 which code the proteins tRNA isopentenyltransferase, Lung seven transmembrane receptor family protein, Ethylene-overproduction protein, Gamma-irradiation and mitomycin c induced 1, putative isoform 3, Protease Do-like 2 ? chloroplastic and Nuclear ribonuclease Z respectively. The remaining 4 epiloci correspond to non-coding regions. In them were found the regulatory elements TATA-box and circadian of S. lycopersicum that could be affecting the expression of nearby genes. These results indicate that DNA methylation in coding and non-coding regions may be causing the observed phenotypic differences between reciprocal hybrids.
