Systemic IL -2/Anti -IL-2Ab Complex Combined with Sublingual Immunotherapy Suppresses Experimental Food Allergy through Induction of Mucosal Tolerance

Resumen:
Cow?s milk allergy(CMA), an immunological mediated reaction to cow?s milk proteins, is one of themost prevalent food allergies in infants and young children. In recent years,it arise again interest in oral and sublingual immunotherapy. Both show promiseas potential therapies. SLIT appears to be safer than the OIT, but its effectivenesshas not yet been fully demonstrated. Recently, it has been demonstrated thatIL-2 complexed with anti-IL-2 monoclonal antibody selective expand regulatory Tcells. We aimed that a combined immunotherapy with low doses of sublingual allergen and lowdoses of the IL-2/anti-IL 2 complex could improve the safety and efficacy ofthe sublingual therapyBalb/c mice weresensitization with cow´s milk protein (CMP) and cholera toxin and then treatedwith PBS (Sens) or CMP (CMPdes) and i.p. injected with IL-2 complex(C or CMPdes/C) Mice were orally challenged with CMP and the immuneresponse was evaluated with in vitro (serum IgE, level; IL-5, IFN-γ secretion by spleen cells and Treg cells, IL-10 andTGF-β in the intestinal mucosa) and in vivo assays (clinical score andcutaneous tests)We found a lowermedium clinical score in treated mice after the oral challenge with CMP, ascompared with sensitized mice, with a reduction in skin mast cell reactivity intreated mice. Immunological changes include decreased specific IgE (2,1±0,3 vs 1,3±0.1CMPdes; 1,4±0,9C; 1,1±0,8CMPdes/C),decreased of Th2 cytokines and induction of IL-10 and Treg in lamina propria ofduodenum (14,6±1,2 vs 28,0±0,6 CMPdes; 35,2±3,1 C; 42,5±1,8 CMPdes/C)and of sublingual mucosa of treated mice (p<0,05). We found that complexadministration induced higher levels of Treg in SLN (p<0,05).
