The Kaposi?s Sarcoma-associated Herpes virus G Protein-CoupledReceptor (vGPCR) is a key molecule in the pathogenesis of Kaposi-Sarcoma. Persistent expression and activity of vGPCR is required for NF-κB pathway activation and tumor maintenance in endothelial cells. The flavonoid quercetin possess potent anti-inflammatory effects due in part to its anti-NF-κB activity, which we supposed to be part of the antiproliferative mecanism. In the present work, we studied the effect of quercetin in the proliferation of vGPCR cells and the regulation ofERK1/2,Aktand NF-κB pathway.Quercetindose response studies (1-20µM)in vGPCR cells showed a reduction on cells number after 24 h treatment. Under contrast phase microscope,cells exhibited apoptotic features asquercetin concentration was increased. Under the same conditions,Western blotstudies showed that incubation of vGPCR cells with quercetin increased IκBαand decreased NF-κB expression. These changes were accompanied by a reduction of p-Aktand p-Erk at low concentration (1-5 µM)of quercetinwhile the opposite effect was observed with higher (10-20 µM)quercetin concentration. All together, these results suggest thatquercetin has antiproliferative and anti-inflammatory effects on endothelial murine cells transformed by vGPCR.
