Effect of different incubation times on tyrosine protein phosphorylation and cleave rate in Iberian red deer thawed spermatozoa

Resumen:
As a first approach, the time of sperm capacitation should be considered.The aim of this work was to study the dynamics of capacitationafter incubation of Iberian red deer thawed spermatozoa with estroussheep serum (ESS) for 1, 5, 15, 30, 45 min and, 1, 2 and 24 hrs. Anegative control without ESS was further evaluated (NCap). Semenof three males was used. Capacitation was assessed by determinationof tyrosine phosphorylation (PY) by western-blot.A heterologousin vitro fertilization was performed according to Soler et al. ReprodDom Anim 43, 293?298 (2008) with capacitated semen 5, 15, 30 and60 min. A significant decrease (p < .05) in the PY values took placein the 34 and 37 kDa at 1, 2 and 24 hrs (1,104 ± 2,471, 520 ± 2,471,613 ± 2,471 and 170,141 ± 132, 104,517 ± 132,132, 9,599 ± 132)compared with 5 and 15 min (10,020 ± 2,471, 10,905 ± 2,471 and632,467 ± 132, 647,640 ± 132). There were differences (p < .05) incleavage rate between 5, 15 and 30 min (20 ± 3, 20 ± 3, 18 ± 3) andNCap and 1 hr of incubation (5 ± 3 and 8 ± 3). In conclusion, PY andcleavage rate exhibit better results under capacitation times lowerthan 30 min in Iberian red deer thawed spermatozoa.
