Fish Type III antifreeze protein (AFPIII) is a prototypical globular antifreeze protein of 7 kDa. It was proposed that, above a critical protein concentration, AFPIII self-assemble to form micelles-like structures that could play a key role on antifreeze activity. In order to understand the complex activities of AFPIII, a comprehensive description of the structural organization in solution is necessary.   Methods We used analytical ultracentrifugation (AUC), size-exclusion chromatography (SEC) coupled to multi-angle laser light scattering (MALLS), and dynamic light scattering (DLS), to characterize the structure and interactions of AFPIII in solution. Small angle neutron scattering (SANS) was used for determining a low-resolution structure of AFPIII in solution. Surface tension of AFPIII in H2O at air/liquid interface was also measured.   Results AUC shows a monodisperse sample of hydrodynamic radius RS=1.6±0.1 nm and molecular weight M=7.3±0.3 kDa for AFPIII, DLS and SEC-MALLS confirm these values respectively. From SANS data  there is no evidence for oligomeric states and the experimental radii of gyration Rg=1.03±0.01 nm agrees with the one from the PDB structure. We also show that AFPIII presents an unusual surface activity.   Conclusion AFPIII at concentrations up to 20 mg/ml is monomeric with a structure in solution compatible with the crystallographic one. Surface tension measurements shows a propensity of AFPIII to localize at the air/water interface, but this surface activity is not correlated with any aggregation in the bulk. We conclude that each AFPIII acts independently of each other, and that specific intermolecular interactions between monomers may not be required prior binding to ice. Complete coverage of ice may not be necesary for antifreeze activity.
