The polyphasic analysis of two native Raphidiopsis isolates supports the unification of the genera Raphidiopsis and Cylindrospermopsis (Nostocales, Cyanobacteria

Raphidiopsis andCylindrospermopsis are planktonic, freshwater bloom-formingcyanobacteria of great concern to human health due to their capability to producepotent cyanotoxins. The presence (in Cylindrospermopsis) or absence (in Raphidiopsis) of heterocysts is the traditionalcharacter used to distinguish between them. This has led to misidentificationsand to question the validity of the genus Raphidiopsis. We studied two R. mediterranea strains isolated from Argentinean shallow lakesusing a polyphasic evaluation that included the morphological description ofthe natural populations, and their ultrastructural, physiological and molecularcharacterization. Heterocyst differentiation was not observed in field or in R. mediterranea cultures submitted tocombined nitrogen deprivation. These results support the occurrence of stablenative R. mediterranea populationswithout heterocysts, which would not be a part of Cylindrospermopsis complex life-cycle. Contrary to earlierpredictions, phylogenetic analysis showed that both strains are more closelyrelated to American Raphidiopsis and C. raciborskii strains than to R. mediterranea and R. curvata strains from other regions, pointing to severalacquisition and loss of heterocysts during evolution. Our results are acontribution to elucidate the controversial taxonomic classification of Raphidiopsis and Cylindrospermopsis since they complement previous reports on thisissue, keeping the question on the validity of the genus Raphidiopsis motivating.
