ABSTRACT Plant roots are colonized by a variety of microorganisms, including the dark septate endophytes (DSE). The aims of this study are to identify and characterize a DSE fungus isolated from wheat roots, characterize and evaluate the natural host response to infection under controlled conditions and perform ultra-anatomical studies. The fungus was isolated and taxonomically identified. Also the isolate was characterized according to their enzymatic and indole production and phosphorus solubilization test. Finally, we performed a greenhouse assay and an ultra-anatomical study. M. bolleyi presented typical DSE structures without neither root tissue alterations nor plant growth effects in different parameters (total dry weight, spike numbers and stem length). The Indol production test showed positive results and TEM observations indicated that the endophyte is able to colonize intercellular spaces without evident plant reaction. The results of this study demonstrate that M. bolleyi doesn´t have negative effects on wheat plants. The response in different environmental conditions and the evaluation of other parameters must be considered in order to establish the importance of this fungus. Key words.
