Doxorubicin loaded magnetic nanotheranostic: Evaluation on the loading capability, their retention during drug targeting and in vivo toxicity

Resumen:
The goal of this work is to evaluate the effect of different reaction parameters on the adsorption of Doxorubicin (Doxo) on magnetitenanoparticles modified with folic acid (MNPs@FA). Three nominal Doxo/MNPs@FA mass ratios, the influence of pH and theincubation time were evaluated to determine the set of parameters ensuring the best formulation regarding, not only the drugloading capability, but also its physicochemical properties. All the obtained samples were characterized by infrared, UV-vis and atomicabsorption (FTIR, UV-vis and AA) spectroscopy, A novel continuous flow based system was designed to estimate their retention abilityin an environment simulating the bloodstream under the influence of an external magnet. This represents a reliable approximationto infer the amount of nanotheranostic able to reach the target site. Besides, in vivo assays were developed on Zebrafish larvae(Danio rerio) model to assess the toxicity of nanotheranostics.
