Resumen:
After human consumption, certain polyphenols can exert cytoprotectiveactions in the gastrointestinal tract, interfering with the oxidativestress implicated in inflammatory disorders. Discards obtained fromthe food industry can be used as important sources of bioactive compounds.In this context,fibremicroparticles (MPCs) obtained fromtheskin of Japanese pulms(Prunussalicina)were evaluatedin the antioxidanteffects of theirpolyphenolsonthecolorectal epithelium (Caco-2)humancells before and after a digestive process.Anin vitrodigestionmethod was carried out with MPCs,accordingto an internationalconsensus standardized static digestion method(Minekus et al2014).Briefly, 1g MCPs was treated withbuffers resemble to gastricandintestinal pH and digestive enzymes (alpha amylase, pepsiny pancreatin).The polyphenoliccomposition(mg/1g)determined inMCPsby HPLCbefore digestionwascyanidin3-galactoside 0.085;cyanidin3-rutinoside 0.036, quercetin3-galactoside + quercetin3-rutinoside0.048; quercetin3-xyloside + quercetin3-rhamnoside 0.036and proanthocyanidns2,000 , while after digestionquercetin3-galactoside+ quercetin3-rutinoside 0.30; quercetin3-xyloside + quercetin3-rhamnoside 0.26; and proanthocyanidins0.33 were determined.Caco-2cell viability was evaluated through MTT assay. Cells treatedfor 24 h with 3 to 100 μg/mlof extract presented a CC50>100 μg/mL.In order to assesst-BOOH induced cellular oxidation through aDCF assay, caco-2 cells were incubated with an ethanolicextract obtainedfrom MCPs or an ethanolicextract fromdigestedMCPs.
