The two isoforms of UGGT are differentially expressed during mouse mammary gland development

UGGT (UDP-Glc::glycoprotein glucosyltransferase) is the key component of the quality control mechanism of glycoprotein folding. It discriminates folded from misfolded glycoproteins and only glucosylates non properly folded proteins that have not reached its native conformation. There are two isoforms of UGGTs (UGGT-1 and UGGT-2) in vertebrates and nematodes. As previous results showed that progesterone regulates the expression and the activity of the two UGGTs isoforms in a mouse hybridoma (a hybrid cell line obtained by fusing antibody producing B cell line and myeloma), we decided to analyze if these enzymes were hormonally regulated during mammary gland development. We determined the level of expression both isoenzymes in C57/BL6 mouse line in different stages: virgin, pregnant, breastfeeding, involution 24 h, involution 48 h, involution 72 h by qPCR and found that both enzyme were differentially regulated. The expression of uggt1 was higher than uggt2 during breastfeeding and mammary involution; but interestingly in virgin and pregnant mice, the uggt2 was at least 2 fold higher than the uggt1.Pregnant mouse have increasing levels of circulating progesterone along pregnancy. We analyzed the expression of both enzymes at day 7, 15 and 17 and results showed a correlation between the level of uggt-2 expression and progesterone level. To study this correlation further, we treated T47D (breast cancer human cell line), for 24 h with increasing concentrations of progesterone (10-8 M, 10-7 M, 10-6 M, 10-5 M) and found the level of uggt2 expression is also regulated by the concentration of progesterone with a maximum of expression at 10-5 M.
