Resumen:
Vasoactive intestinal pepetide (VIP) is produce by trophoblast cells and displays multiple target circuits that allow immunetolerance to pregnancy-related antigens, however, its interaction with progesterone (P4) in this system is still unknown. Objetive: We evaluated the immunomodulatory effects of P4 and VIP on trophoblast cell function focusing on their pahgocytic ability. Methods: We used an in vitro model based on the Swan71 cell line cocultured or not with PBMC from fertile women and we evaluated nitrites (by Griess Method), MMP9 (by Zymography), ROS production by FACS, COX-2 and VPACs expression (by RT-PCR). Phagocytic ability was quantified usign latex-beads FITC-conjugated (efferocytosis) or apoptotic red blood cells CFSE-stained (eryptosis) by FACS and the cytokines production by RT-PCR. Results: After the interaction between Swan cells and maternal PBMC in the presence of VIP (10-7M) and P4 (10-4M) nitrites production, MMP9 activity and COX-2 expression decreased (p
