Immune responses induced upon infection of human monocytes from tuberculosis patients with live Mycobacterium tuberculosis strains.

Resumen:
Background: A deeper comprehension ofMycobacterium tuberculosis(Mtb) pathogenesis is essential forsetting new preventive strategies. Mycobacterium RD1-locusencodes a specialized secretion system and virulence effectorproteins that contributes to mycobacterial regulation of hostimmune responses. Autophagy was shown to display a critical roleduring human immune response against Mtb. Actually; autophagyinduction promotes maturation of Mtb-containing phagosomes,suppressing mycobacterial survival. Here we investigated themodulation of the autophagy process and the production ofcytokines in monocytes infected with a RD1 deleted-Mtbstrain(MtbdeltaRD1),or MtbH37Rv pathogenic strain.Methods:Peripheral blood mononuclear cells(PBMCs) fromhealthy donors(HD) and tuberculosis patients(TB) were eitherinfected with MtbH37Rv or MtbdeltaRD1, or stimulated withsonicatedMtb(Mtb-Ag), ±bafilomycinA1(BafA1). After24h, wedetermined the autophagy levels(LC3-II) by Western Blot andImmunofluorescence microscopy. IFN-gamma and TNF-alphalevels were analyzed byELISA.Results:In contrast to MtbH37Rv, a strain known to interfere withthe phagosome maturation pathway, after infection withMtbdeltaRD1, monocytes displayed high levels of LC3-IIB/puncta. Thus, MtbdeltaRD1strain was able to induce afunctional autophagic process, as evidenced by a significantincreased of LC3-IIBpuncta/cell in the presence ofBafA1(p
