Melatonin (MEL), hormone involved in circadian rhythms synchronization, also acts as an antioxidant, immune modulator and promotes bone and fibroblastic formation. Since salivary and serum MEL levels decrease in periodontitis, MEL is proposed as a therapeutic agent for oral cavity.AIM: To investigate the effects of MEL on growing fibroblast (FB) culture from rat gingiva and to compare it with other agents. METHODS: A primary culture of maxillary gingiva from Wistar rats in enriched culture medium (CM) was performed. After reaching cell confluence, FB were plated (FB 45000 ml/well) and divided into 3 experimental groups: 1) Control (FB-MC only), 2) FB+collagen(FB-MC + lysate collagen 0.01%) 3) FB+MEL (FB-MC + solution MEL 50 nM). To perform the growth curve, cells were suspended with a solution of trypsin (0.05%) and counted with the Neubauer chamber and crystal violet staining for 9 days. ANOVA with Bonferroni comparisons for analysis were used. RESULTS we found that between 3 and 6 day of culture, FB showed exponential growth.
