Influenceof Gonadotropin-releasing hormone antagonistcetrorelix from celiac ganglion on theovarian estradiol and nitric oxide release.There are background demonstrating the extra hypothalamic GnRH source as well as an inhibitory effect on ovarian steroidogenesis. Taking into account this information, the aims of this work was to study the effect of a GnRH antagonist on: 1) the ovarian release of estradiol (E2) and nitric oxide (NO), 2) ovarian gene expression of P450aromatase, E2 synthesis enzyme, endothelial nitric oxide synthase (eNOS) and inducible nitricoxide synthase (iNOS) on day 21 of pregnancy in the rat using the ex vivo CG-SON-ovary system. The system was incubated in Krebs Ringer at 37ºC, keeping CG and ovary connected by the SON, in separate compartments. The GnRH antagonist: cetrorelix 10-6 M (Ctx) was added in the ganglion compartment. E2 was determined by RIA and NO by the Griess technique in the ovarian compartment at 30 and 180 min of incubation. Ovarian gene expressions of aromatase, eNOS and iNOS were assessed by RT-PCR at 180min. One-way ANOVA and Tukey testwere used (p<0.05).Ctx added in CG increased the E2 and NO release significantly at 180 min (p<0.01 and p<0.05, respectively) without modify the expression enzymes analyzed. In conclusion, cetrorelix from CG stimulate the ovarian estradiolrelease being the NO the possible messenger molecule involved.
