Resumen:
ANDROSTENEDIONE LUTEOTROPHIC ACTION, THROUGH THE NEURAL PATHWAY, IS MODIFIED BY FLUTAMIDE IN COELIAC GANGLION. In he coeliac ganglion-superior ovarian nerve-ovary (CG-SON-Ovary) system, we have shown that at the begining of the functional luteal regression (pregnancy day 19), androstenedione (A2),  stimulates the liberation of luteal progesterone (P) through the neural pathway. Here we studied if the ganglionic action of A2 is mediated by androgen receptor and if that action is modified by ganglionic sympathetic stimulation. The CG-SON-Ovary system was incubated in Krebs Ringer-glucose-albumin (0.1 mg/ml) at 37ºC, keeping CG and ovary connected by the SON, in separate cuvettes. In the CG compartment was added A2 or NA (10-6M), flutamide- androgen receptor antagonist (10-4M) [(A2)g, (NA)g, (Flu)g respectively] and the combinations [(Flu+A2)g, (NA+A2)g (NA+Flu)g, (A2 +Flu+NA)g]. Controls were not stimulated. The P liberated was measured (by RIA) in the ovary compartment at 30, 60 and 120 min of incubation system. Results were: Control P (ng/mg ovary; mean ±S.E.M) at 30?: 0.20±0.02; 60?: 0.22±0.03; 120?: 0.22±0.03. Control vs (Flu)g: P release was not modified. (A2)g vs (Flu)g and (NA+A2)g vs (A2 +Flu+NA)g: P release was decreased at all times (p<0.001and p<0.025 respectively). Thus,  flutamide has no per se effect in ganglion but modifies the A2 effect and the neuroendocrine action (NA+ A2) suggesting the involvement of CG androgen receptors in the A2 effect on the P release.
