Previous studies carried out in ourlaboratory suggests that L. tenuis (Lt) and L. corniculatus (Lc) arean invaluable tool to study the genetic control of proanthocyanidins (PAs)biosynthesis and tolerance to stresses in legumes. With these aim we performedan RNA-Seq sequencing to obtain the full-length transcriptome of both species.For these, RNA samples were isolated from shoots of a diploid accession of Lc and Lt (n=3) and were used to performed a high-performancepaired-end (2x100bp) sequencing on Hiseq 1500-Illumina. From obtained reads weperformed two different analyses: a) de novo assembly using Trinity programand b) assembling and mapping to L. japonicus genome using TopHat andCufflinks programs. From a)analysis, we obtained 91,530 and 80,911 genes for Lc and Lt respectively,functional annotation of these genes was performed using Trinotate pipeline. Onthe other hand, between 56 to 59% of transcripts were mapped against L. japonicus genome from b) analysis; allowing theidentification of 42,013 genes. From these, 12,868 genes were differentially expressed between Lc and Lt, and 4,912 of those had fold change ≥|2|. GO enrichment analysisby Blast2GO of differentially expressed genes was performed to determinate the GO terms highly represented. Thiswork represents the first report of full transcriptome of L. tenuis and a diploid L.corniculatus, raising the possibility of future agricultural studies andapplications in the others forage legumes.
