Granulocyte-Macrofage Colony Stimulating Factor (GM-CSF) is a cytokine that regulates the proliferation and differentiation of hematopoietic progenitor cells, useful to reverse or prevent chemotheraphy secondary leukopenias. It was expressed in different host systems: bacteria, yeast and mammalian cells. All of them have biological activity. However, patients receiving non-glycosylated or partially glycosylated GM CSF develope antibodies that compromise the clinical efficacy of the trial (1). To study the undesirable immune response we produced a panel of E. coli derived recombinant human GM CSF (rhGM CSF) monoclonal antibodies (MAbs). We characterized the MAb´s immunochemical behaviour and neutralizing capacity in order to use this panel for evaluating the GM CSF epitopes involved in the therapeutic failure in humans.
