Ce-TiO2 material with promising characteristics for osseointegration: Interaction with fibroblasts and effect of albumin adsorption and bioactivity on 3D surface topography

Resumen:
The success of dental and bone implants strongly depends on a good integration of the biomaterial to the surrounding tissues. This is highly affected by the characteristics of the implant surface which has to allow adsorption of certain proteins, cell anchorage and hydroxyapatite deposition. In the present study we used a microemulsion route to develop a novel material based on cerium doped TiO2 and evaluated the properties mentioned above.Mouse fibroblasts viability has been measured after 72 h of co-incubation with increasing concentrations of the particulate material (from 10 to 1000 μg/mL) showing no statistical differences in growth respect to untreated cells (p > 0.10). Morphology using phase contrast microscopy was also evaluated with similar results.Bioactivity, this is the ability of the material to stimulate the deposition of hydroxyapatite on its surface after soaking in Simulated Body Fluid (SBF), was investigated in presence of a pre-adsorbed layer of Bovine Serum Albumin (BSA). SEM images show the development of a thick layer of Ca?P deposits of plate-like crystals disposed in a rosette-like shape. Moreover, roughness parameters are preserved after SBF treatment (Rku = 1.654 ± 0.043; Rsk = 1.236 ± 0.014) with respect to material's original topography (Rku = 2.404 ± 0.169; Rsk = 1.466 ± 0.034).Positive values of skewness (Rsk) and kurtosis (Rku) magnitudes ≤ 3, are generally regarded as improvers of interface shear strength which, added to its bioactivity in presence of BSA and biocompatibility with fibroblasts, become favorable features for osseointegration of the implant.
