Recombinant human growth hormone (rhGH) is used forthe treatment of several pathologies, most of them related togrowth. Although there are several expression systems usedfor its production, the milk from transgenic cows is one of themost interesting due to the high levels of rhGH achieved (5 g/l).We have designed and synthesized a short peptide using Fmocchemistry and studied its ability to purify rhGH from milk onceimmobilized on a Sepharose support. The adsorption isothermfor the rhGH-peptide system showed a dissociation constant of3.1 × 10-6 M, more moderate than those of antibodies, whichare in the order of 10-9 M. Once performed the chromatographybetween our generated peptide-resin and spiked milk withrhGH, we analyzed with SDS-PAGE the wash and elutionfractions and observed that rhGH was purified with over 90%purity in a single step. This proves that short peptides are anefficient and economical tool for the purification of therapeuticproteins from complex mixtures.
