Thyroid hormones have a key role in the tuning of mitochondrial respiratory rate, although the involvedmechanisms are not completed defined yet. In the present work, it is tested whether nitric oxide released by livermitochondrial nitric oxide synthase (mtNOS) participates in the modulation of oxygen uptake in thyroid disorders.Mt NOS expression and activity were selectively increased in hypothyroidism (2-3 fold) and reverted by hormonereplacement. No significant changes in other isoforms were observed. MtNOS activity correlated with serum T3levels (r: -0.73, p= 0.003), and systemic and mitochondrial oxygen uptake (r:-0.77, p<0.0001). Moreover, mitochondrial oxygen uptake and cytochrome oxidase activity were further decreased in the presence ofL-arginine (0.1-0.3 mM) which is consistent with 5-10 fold increase in matrical NO concentration. These resultssupport the notion that thyroid-related changes in mtNOS and in the resulted matrical NO concentration representa substantial part of the oxygen uptake regulation and that this causative biochemical mechanism could beextended to other tissues and adaptive conditions associated to changes in thyroid hormone tissue levels.
