On the other hand, polyphenols from grape (bioactive compounds of plant origin with antioxidant activity) has been found to exert anti-inflammatory effect on adipose tissue. The aim of this study was to evaluate the effect of malbec grape pomace extract (GPE) on adipose tissue angiogenesis/adipogenesis and inflammation from Wistar-Kyoto (WKY) rats receiving a high-fat diet (HFD). Ten weeks old WKY (n=16) rats were divided into three groups: Control group WKY (n=4); WKY-HFD group receiving fat (40% (w/w), (n=6); and WKY- HFD supplemented with GPE (300 mg/Kg/d, n=6) for 10 weeks. HFD increased parameters of metabolic syndrome: body weight gain, systolic blood pressure, triglycerides and decreased HDL-cholesterol that were ameliorated/prevented by GPE. Also, HFD leads to an increased epididymal adipose weight and adipocytes diameter size accompanied with high levels of free fatty acid (FFA). GPE supplementation decreased plasma levels of FFA and adipocyte size without changes in adipose weight. Moreover, GPE increased adipose tissue angiogenesis in WKY rats evidenced by the expression of vascular endothelial growth factor A (VEGF-A), and its receptor VEGFR-2. These changes were associated with an increased adipogénesis, (PPAR-y) protein expression observed in WKY- GPE rats compared with WKY-HFD. HFD increased parameters of adipose tissue inflammation CD68, Nox4 and resistin that were attenuated by the supplementation with GPE. Considering these results, the increase angiogenesis and adipogenesis of adipose tissue would give rise to a cell number increase (hyperplasia of the tissue), decreasing therefore the size of the adipocyte and producing a decrease adipose tissue inflammation. Overall, supplementation with food rich in bioactive compounds can attenuate adipose tissue inflammation perhaps by its effect on adipose tissue angiogenesis and adipogenesis.
