Resumen:
This studyaimedtodeterminewhethertheeffectsonthemouselivercausedbythreeperiodsoffeeding
a protein-freedietfor5daysfollowedbyanormalcompletedietfor5days(3PFD-CD)arepreventedby
a constantmethioninesupply(3PFD+Met-CD).TheexpressionsofcarbonicanhydraseIII(CAIII),fatty
acid synthase(FAS),glyceraldehyde3-phosphatedehydrogenase(GAPDH)andglutathioneS-transferase
P1 (GSTP1)wereassessedbyproteomicsandreversetranscriptase-polymerasechainreactions.Theliver
redox statuswasexaminedbymeasuringtheactivitiesofsuperoxidedismutase(SOD)andcatalase(CAT),
as wellasproteincarbonylation.Becauseoxidativestresscanresultinapoptosis,theactivityandcon-
tent ofcaspase-3,aswellasthex-linkedinhibitoroftheapoptosisprotein(XIAP)andmitochondrial
caspase-independent apoptosisinducingfactor(AIF)contentswereassessed.Inaddition,theliverhis-
tomorphology wasexamined.Comparedtothecontrolsfedanormalcompletedietthroughout,feeding
with 3PFD-CDincreasedtheFAScontent,decreasedtheCAIIIcontent,decreasedboththeSODandCAT
activities, andincreasedproteincarbonylation.Italsoactivatedcaspase-3,decreasedtheXIAPcontent,
decreased theAIFcontent,increasedthenumberofGSTP1-positivefociandcaspase-3-positivecells,and
caused fattylivers.Conversely,thechangeswerelessenedtovaryingdegreesinmicefed3PFD+Met-CD.
The presentresultsindicatethataregularMetsupplylessensthebiochemicalchanges,damage,and
caspase-dependent apoptosisprovokedbyrecurrentdietaryaminoaciddeprivationinthemouseliver
